RESUMO

The development of speech is a process by which the neural network structure of brain perceives, decodes, represents, and outputs input signals. At the cortical level, the classic model of speech processing proposed two pathways leading from the auditory system: the ventral stream and the dorsal stream.The ventral stream, which involved structures in the superior and middle portions of the temporal lobe, was involved in processing speech signals for perception and comprehension (speech recognition). The dorsal stream, which involved structures in the posterior frontal lobe and the posterior dorsal-most aspect of the temporal lobe and parietal operculum, had an auditory-motor integration function and was essential for speech production. However, the importance of auditory processing in central auditory nervous system(CANS) from the vestibulocochlear nerve to the auditory cortex for speech development had not been clarified.This review aims to discuss the roles and related mechanisms of the CANS in speech recognition and generation from the perspective of cognitive neuroscience, so as to provide new ideas for clinical monitoring and long-term prediction of auditory processing and speech development, and for diagnosis and treatment of clinical speech-related diseases.

Objective To explore the correlation between serum lipoprotein-related phospholipase A2 (Lp-PLA2), cystatin C (Cys-C) levels and disease severity in patients with hepatitis B cirrhosis (HBC). Methods Clinical data of 110 HBC patients in the hospital from October 2017 to May 2019 were retrospectively analyzed. According to Child-Pugh classification criteria of liver cirrhosis, they were divided into grade A (n=42), grade B (n=37) and grade C groups (n=31). Another 30 healthy controls during the same period were enrolled as control group. The levels of serum Lp-PLA2 and Cys-C were detected. And their correlation with disease severity was analyzed. Results Levels of serum Lp-PLA2 and Cys-C in HBC group were higher than those in control group (P<0.05). The levels of serum Lp-PLA2 and Cys-C were the highest in grade C group, followed by grade B group and grade A group (P<0.05). The areas under the ROC curve (AUC) of serum Lp-PLA2 combined with Cys-C for evaluating grade A and B, grade B and C HBC were 0.875 and 0.837, which were higher than those of Lp-PLA2 (0.772, 0.750) and Cys-C (0.750, 0.691) alone (P<0.05). Spearmann rank correlation analysis showed that levels of serum Lp-PLA2 and Cys-C were positively correlated with disease severity (r=0.659, 0.561, P<0.05). Conclusion The levels of serum Lp-PLA2 and Cys-C are significantly increased in HBC patients, which are gradually increased with the aggravation of HBC. The two indexes are positively correlated with disease severity, which are of diagnostic efficiency for the classification of liver cirrhosis.

Objective: To investigate the effect of CXCL12/CXCR4/CXCR7 axis on the metastasis and invasion in pancreatic cancer so as to provide new evidence for research on pancreatic cancer metastasis treatment. Methods: MiaPaCa-2 cells were transfected with CXCR4 shRNA and CXCR7 shRNA, and the Transwell assay was used to determine the effects of CXCL12/CXCR4/CXCR7 axis on cell invasion and migration. Quantitative RT-PCR and Western blotting were used to explore the effects of CXCL12/CXCR4/CXCR7 axis on the expressions of invasion-related genes (MMP-2 and uPA) and EMT-related genes (E-cadherin and Vimentin). Results: CXCL12 significantly increased the metastasis and invasion of pancreatic cancer cells. The enhancement of tumor cell invasion was effectively countered by CXCR4 shRNA or CXCR7 shRNA. CXCL12/CXCR4 axis in cancer cells increased the expressions of invasion-related genes (MMP-2 and uPA) and EMT-related genes (E-cadherin and Vimentin). CXCL12/CXCR7 axis only increased the expressions of MMP-2 and uPA. Compared to blocking CXCR4 or CXCR7 alone, the inhibitory effects on invasion-related genes and EMT-related genes were more effective when both CXCR4 and CXCR7 were blocked. Conclusion: CXCL12/CXCR4/CXCR7 axis regulates the EMT, metastasis, and invasion of pancreatic cancer cells.

Objective To explore the effect of early enteral nutrition (EEN) and parenteral nutrition (PN) on the postoperative outcomes of patients with gastric cancer and nutritional risk in enhanced recovery after surgery. Methods A total of 130 patients with gastric cancer hospitalized in department of surgery of Ningbo First Hospitalfrom September 2016 to May 2018 were selected and divided into early enteral nutrition support group (EEN) which was placed with jejunal nutrition tube during the operation, and enteral nutrition started within 12-24 hours after the operation, and parenteral nutrition support group (PN) which was given parenteral nutrition support one day after surgery. Patients in both groups were given nutrients of equal heat and nitrogen. The incidence of nutrition-related complications, the incidence of infection-related complications, the length of postoperative hospital stay and the time of anal exhaust were compared between the two groups. Results The incidence of nutrition-related complications was 10 cases (15. 38%) and 4 cases (6. 15%) in EEN group and PN group, that was not statistically different (P = 0. 157). The incidence of infection-related complications was 3 cases (4. 61%) and 5 cases (7. 69%) in EEN group and PN group, that was not statistically different (P = 0. 715). The postoperative hospital stay was 11 days (range, 10-15) and 12 days (range, 11-13) in EEN group and PN group, that was not statistically different (P = 0. 233). The first anal exhaust time and defecation timewere 64 hours (range, 52-77) and 87 hours (range, 76-100) in EEN group and 72 hours (range, 60-86) and 96 hours (range, 86-120) in PN group, that was statistically different (P=0. 001, P=0. 034). Conclusion Enhanced recovery after surgery, early enteral nutrition after gastric cancer surgery may promote the recovery of intestinal function, but the complications and hospital stay after operation are not improved.

The NAC family is an important transcription factor which regulate plant growth and development, signal transduction, and stress response.In this study, the protein identification, subfamily classification, the determination of physical and chemical properties, protein structure, and expression pattern of NAC family were performed using bioinformatic methods based on the RNA-seq data of ginger. The results showed that a total of 72 NAC transcription factors were identified in 271.1 Mb total nucleotides, and they could be clustered into 13 subfamilies according to the phylogenetic tree.The physical and chemical properties, structure analysis revealed that the amino acid number and isoelectric point were different among 13 NAC subfamilies; the secondary structure of NACs transcription factors mainly consist of random coil, and the tertiary structure is similar.In addition,the expression patterns of genes under different soil moisture and Ralstonia solanacearum infection showed that 23 NACs were differentially expressed, which were mainly distributed in Ⅷ,Ⅶ, and ⅩⅤ subfamilies related to plant senescence, hormone metabolism and cell wall metabolism.The results provide some valuable information for the research and development of NAC transcription factors in ginger.

Objective To study the effect of [Gly14]-Humanin on oxidative stress and neuron apoptosis after focal cerebral ischemia reperfusion injury in rats. Methods A rat model of acute middle cerebral artery occlusion(MCAO)and reperfusion was established by suture embolism. Ninety-six healthy male Sprague-Dawley rats were randomly divided into sham-operation group, model group, normal saline group and HNG group. And 5 μL HNG(100 nmol/L)in the rats of the HNG group, normal saline in the rats of the sham-operation group and normal saline group were given to rats 3 days before operation respectively(iv, three times, qd). After cerebral ischemia 3 h and 24 h of reperfusion, neurological deficit score(NDS) were performed for each group, and the activity of sod, levels of GSH and MDA in cerebral tissue were detected. TUNEL staining were used to detect neuronal apoptosis. Results Median(Interquartile range)of NDS was 0(0)in sham-operation group, 1.5(1)in model group, 3(1)in normal saline group and 3(1)in HNG group, respectively, and the NDS of sham-operation group was significantly lower than the other three groups. The NDS of HNG group was significantly lower than the model group and normal saline group(all P<0.05). Compared with sham-operation group, the activity of SOD of model group, normal saline group and HNG group rats[(11.65±1.66),(12.15±1.56)and (19.43±1.47)U/mL]and levels of GSH[(8.84±1.23),(7.51±1.16)and (11.17±1.67)mg/L]were decreased, and levels of MDA[(42.61±1.79), (40.33±1.24)and (35.39±1.29)nmol/mL]were increased(P<0.05). The activity of SOD and levels of GSH of HNG group were significantly higher than model group and normal saline group, but the levels of MDA was significantly lower than model group and normal saline group(all P<0.05). Compared with sham-operation group[(0.13±0.01)%], percentage of neuronal apoptosis of model group, normal saline group and HNG group rats[(0.59 ±0.07)%,(0.51 ±0.08)% and (0.22 ±0.03)%, respectively]were increased(P<0.05), and percentage of neuronal apoptosis of HNG group was significantly lower than model group and normal saline group(all P<0.05). Conclusion By increasing the activity of SOD and levels of GSH,[Gly14]-Humanin can resist focal cerebral ischemia reperfusion injury, and can decrease the neuro apoptosis of ischemic area, thus mitigate the neurologic deficit.

AIM:To investigate the role of zinc finger protein 281(ZNF281)in the proliferation of hepatocel-lular carcinoma(HCC)cells.METHODS:The mRNA expression levels of ZNF281 in peripheral blood mononuclear cells from 80 cases of healthy people and 206 cases of HCC patients were determined by real-time PCR.Statistical analysis were used to illustrate the relationship between the mRNA expression levels of ZNF 281 in the peripheral blood mononuclear cells and the clinicopathologic parameters of HCC patients.Real-time PCR and Western blot were used to detect the mRNA and protein expression levels of ZNF281 in hepatoma cell lines and immortalized hepatocytes.The silencing of ZNF281 was conducted by transfection of small interfering RNA targeting ZNF281,and then the proliferation of HCC cells was analyzed by MTS assay.The DNA synthesis of HCC cells was tested by Cell-LightTMEdU Apollo?488 In Vitro Imaging Kit.The ability of colony formation of the HCC cells was measured by colony formation assay,and the ability of anchorage-indepen-dent growth was detected by soft agar test.RESULTS: The mRNA expression level of ZNF281 in the peripheral blood mononuclear cells from HCC patients was significantly increased compared with the healthy people,and the high expression level was positively correlated with tumor size,tumor stage and tumor vascular invasion.Concurrently,the expression level of ZNF281 in hepatoma cell lines was significantly higher than that in immortalized hepatocytes.More importantly,the si-lencing of ZNF281 inhibited the proliferation, DNA synthesis, colony formation and anchorage-independent growth of the HCC cells.CONCLUSION:ZNF281 promotes the proliferation of HCC cells.

Objective To develop norms for the rapid automatic naming and rapid alternating stimulus tests (RAN/RAS tests)on school children.Methods A multi-stage sampling procedure was used to select primary school students randomly from 8 schools in Nanjing,aged from 7 to 12 years old.The RAN/RAS tests included 6 classes of stimuli:objects,colors,numbers,simplified Chinese characters,numbers and simplified Chinese characters mixture,numbers and simplified Chinese characters and colors mixture. Results (1)A total of 1 048 students were randomly selected to participate in the study in 6 groups,with sex ratio of 1.001.04.(2)The scores of the boys and girls did not differ sig-nificantly by any of the methods(objects,colors,numbers,simplified Chinese characters,numbers and simplified Chinese characters mixture,numbers and simplified Chinese characters and colors mixture)(all P>0.05).(3)On any of the measures in all groups,the scores decreased as the age increasing(P<0.001).(4)Compared with the three-item scores (objects,colors,numbers)between Chinese and English,there were significant statistical differences among all age groups (all P<0.001).(5)Time reliability correlation coefficient ranged from 0.84 to 0.92,and retest reliability correlation coefficient ranged from 0.98 to 0.99.The validity test showed that the correlation coefficient was statistically significant (P<0.05).Conclusions The scores of RAN/RAS tests norms have nice reliability and validity and are in line with the law of development.The norms derived in this study are recommended as the norm RAN/RAS scores for Chinese children.

Objective To explore the clinical effects of oxytocin intravenous drip combined with psychological intervention on the treatment of labor induction. Methods 98 patients with odinopoeia were andomly divided into the study group (n=49) and the control group (n=49). The control group were given oxytocin intravenous drop, the study group were received intravenous infusion of oxytocin combined with psychological intervention. Mode of delivery in the two groups were recorded and the effects were compared. Results Nobody exit in this study. There was no withdrawal, interruption, death and other abnormalities in this study. The natural childbirth rate of the study group was 77.55%, the natural birth rate of the control group was 55.10%. The cesarean section rate was 18.37% in the study group and 55.10% in the control group, and the difference between the 2 groups was statistically significant (P<0.05). There had no statistically significant difference in the rate of forceps delivery between the two groups. Conclusion Oxytocin intravenous drip combined with psychological intervention applied to odinopoeia can significantly improve the natural childbirth rate .

Objective @#To explore the method of apically positioned flap technique (APFT) on buccal keratinized gingiva reconstruction around dental implants and evaluate its clinical outcomes and technical characters. @*Methods @#13 patients, who were insufficient of buccal keratinized gingiva around dental implants but sufficient with alveolar ridge crest tissue or palatal tissue at posterior maxillar, were enrolled in this study. APFT was operated during the phase Ⅱ surgery, by which some tissues were transferred from the alveolar ridge crest or palatal side to the original site of keratinized gingiva. Before APFT surgery and 1, 6 and 12 months after crown restoration, the width and thickness of transferred tissues were measured, gingival index (GI) and bleeding on probing (BOP) were also detected. The results were applied to comparative t-test statistical analysis.@*Results @# Transferred tissues by APFT showed healthy one month after crown restoration and exhibited characters of keratinized gingiva compared with the adjacent teeth at 6 and 12 months after restoration. Mean value of width of transferred tissue were respectively (3.25 ± 0.40) mm, (3.04 ± 0.34) mm and (2.97 ± 0.32) mm, meanwhile the thickness were respectively (2.05 ± 0.20) mm, (1.91 ± 0.23) mm and (1.84 ± 0.25) mm. The value of width and thickness of the adjacent teeth gingiva were (3.19 ± 0.42) mm and (1.96 ± 0.23) mm respectively. No significant differences were found between transferred tissue and adjacent teeth gingiva on width and thickness (P>0.05). Observation results of GI and positive rate of BOP of transferred tissue were also similar to which of gingiva of the adjacent teeth. @*Conclusion @#Technique of apically positioned flap is an effective measure on buccal keratinized gingiva reconstruction.

Objective To explore the endoscopic characteristics of multidrug-resistant tuberculosis (MDR-TB) combined with tracheobronchial tuberculosis (TBTB). Methods 248 MDR-TB as study group, they hospitalized from October 1st 2008 to June 31st, 2016. 274 cases of non MDR-TB with bacteria positive as control group over 2015, all of them received bronchoscopy, sputum cultured and drug sensitivity tested of Isoniazid and Rifampicin. We analyzed the results of bronchoscopy and demographic data. Results 248 cases of MDR-TB patients, of 175 (70.56%) were diagnosed TBTB by bronchoscopy, of 73 (29.44%) without TBTB. 274 cases of non MDR-TB with bacteria positive patients, of 146 (53.28%) were diagnosed TBTB, of 128 (46.72%) non TBTB, the difference of comparisons was statistically significant (χ2 = 16.42, P = 0.000). MDR-TB combined with TBTB median age was 32 years, non MDR-TB combined with TBTB median age 42 years, the difference was statistically significant (U = 9932.00, P = 0.001). Among the MDR-TB patients, of 75 (42.86%) TBTB in the upper right bronchial, of71 (40.57%) upper left bronchus, while non MDR-TB patients, of 70 (47.95%) and 60 (41.10%), there was no statistically significant difference (χ2 = 2.44, P = 0.786). Among the MDR-TB, of 76 (43.43%) were inflammation infiltration type, of 11 (6.29%) were necrosis type, of 13 cases (7.43%) granulation proliferative type, of 72 (41.14%) were scar stricture type, of 3 (1.71%) tube wall softening type. Among the non MDR-TB, in turn, TBTB type were 50 (34.25%), 41 (28.08%), 9 (6.16%), 40 (27.40%), 5 (3.43%), the difference were statistically significant (χ2 = 30.50, P = 0.000). Conclusions The detection rate of TBTB was higher in MDR-TB patients, that common occur in younger patients. TBTB common infringe on upper right bronchial and upper left bronchus, TBTB type most are inflammatory infiltration type and scar stricture type. More attention should be paid to bronchoscopy among MDR-TB patients.

<p><b>OBJECTIVE</b>To explore the effect and action mechanism of moxibustion on healing of cutaneous wound in rats.</p><p><b>METHODS</b>Twenty-four SD rats were selected and made into linear full-thickness skin injury model. With randomized digital table, rats were randomly divided into a treatment group and a model group, 12 cases in each one. Then according to treatment time, each group was again divided into a 1d group, a 3d group and a 7d group, 4 cases in each one. The moxibustion at injured skin was applied in the treatment group, 30 min per time, once a day. Hematoxylineosin (HE) staining method was adapted to measure growth status of capillary and number of vascular endothelial cell; immunohistochemical method was used to measure the expression of vascular endothelial growth factor (VEGF).</p><p><b>RESULTS</b>The wound healing indices in the treatment 7d group were higher than those in the model 7d group on both the 4th day and 8th day after treatment (both P < 0.05). The number of capillary in the treatment 1d group and 3d group was higher than that in the model 1 d and 3 d groups (both 1 < 0.05). The number of capillary in the treatment 7d group was lower than that in the model 7d group (P < 0.05). The number of vascular endothelial cell in the treatment 3d group was higher than that in the model 3d group (P < 0.05). The number of vascular endothelial cell in the treatment 7d group was lower than that in the model 7d group (P < 0.05). The difference of number of vascular endothelial cell between the treatment 1d group and model 1d group was not significant (P > 0.05). Positive cells accumulated score of V EGF expression in the treatment 3d group was higher than that in the model 3d group (P < 0.05). Positive cells accumulated score of VEGF expression in the treatment 7d group was lower than that in the model 7d group (P < 0.05). The difference of positive cells accumulated score of VEGF expression between the treatment 1d group and model 1d group was not significant (P > 0.05).</p><p><b>CONCLUSION</b>Moxibustion could improve the healing of skin wound in rats, which could be related with regulating vascular endothelial cell and VEGF in wound tissue at different time.</p>

<p><b>OBJECTIVE</b>To investigate the relationships between the severity of childhood allergic rhinitis (AR) and the peripheral blood eosinophil count, serum eosinophil cationic protein (ECP), total IgE (tIgE), and allergen-specific IgE (sIgE) levels.</p><p><b>METHODS</b>A total of 138 children with AR aged 3 to 17 (9.96 ± 3.78, x() ± s) years old were enrolled in the study. All children had persistent AR sensitized to house dust mites with a clinical history of 3 months to 12 (4.21 ± 2.72) years. The disease severity was evaluated using 10 cm-visual analogue scale (VAS), and the serum levels of ECP, tIgE and sIgE were determined using an ImmunoCAP system. Statistical analysis was conducted with SPSS11.0 software.</p><p><b>RESULTS</b>Among 138 children with AR, the VAS scores for global severity of rhinitis and nasal obstruction symptom were 5.32 ± 2.16 and 4.78 ± 2.45, respectively. Blood eosinophil count was 0.39 [0.24; 0.63] (M[P(25); P(75)]) ×10(9)/ml. Serum levels of ECP and total IgE were 10.60 [3.26; 30.80] µg/L and (2.50 ± 0.53) log kU/L, respectively. Serum levels of allergen-sIgE against Dermatophagoides pteronyssinus and Dermatophagoides farinae were 58.20[24.75; > 100] kUA/L and 54.95 [24.55; > 100] kUA/L, respectively. The VAS scores of nasal obstruction symptom, but not global severity of rhinitis, were positively related to the duration of AR (r = 0.215, P = 0.011) and the levels of serum ECP (r = 0.196, P = 0.022) in bivariate correlation analysis. There was also a significant correlation between the serum ECP level and the blood eosinophil count (r = 0.295, P = 0.000). No relationships of blood eosinophil count, and serum tIgE and sIgE levels with global severity of rhinitis as well as nasal obstruction symptom were found (all P > 0.05).</p><p><b>CONCLUSIONS</b>These results suggested that the severity of nasal obstruction was positively correlated with the duration of rhinitis and the levels of serum ECP in childhood persistent AR due to house dust mites, indicating the disease severity might be related to chronic inflammatory process.</p>

<p><b>OBJECTIVE</b>To study the growth rhythm of preterm appropriate for gestational age (AGA) infants by investigating their physical catch-up growth characteristics.</p><p><b>METHODS</b>Eighty-four preterm AGA newborns (44 males and 40 females) with gestational ages between 28-36 weeks were enrolled. The weight, length and head circumference were evaluated by Z score according to the criterions of actual and corrected ages.</p><p><b>RESULTS</b>The preterm infants had the catch-up growth in weight, length and head circumference in the first year of life. The growth velocity within the first three months was the highest. The velocity in the weight catch-up was higher than that in the length.</p><p><b>CONCLUSIONS</b>The fastest growing period is the early three month of life in preterm AGA infants. The catch-up growth in weight and length is unbalanced.</p>

<p><b>OBJECTIVE</b>To investigate the association between the promoter polymorphism -509C/T of the transforming growth factor-β1 gene (TGFB1) and the disease severity of allergic rhinitis (AR) in childhood.</p><p><b>METHODS</b>A total of 96 Chinese patients with persistent AR aged 3 - 17 (9.4 ± 3.8) years old were enrolled in the study. Among these patients 53.1% were mild cases (n = 51) and 46.9% were moderate-to-severe cases (n = 45). Genotyping was performed on peripheral blood genomic DNA by using PCR-RFLP. Serum levels of TGF-β1 was measured by ELISA, and serum total IgE, specific IgE and eosinophil cationic protein (ECP) levels were determined using an ImmunoCAP100E system. Statistical analysis was conducted with SPSS11.0 software.</p><p><b>RESULTS</b>Significant differences were found in genotype frequencies for the TGFB1-509C/T polymorphism between mild and moderate-to-severe AR patients (χ(2) = 8.361, P = 0.015). Children with persistent AR bearing the TT genotype of the -509C/T polymorphism had significantly increased risk for moderate-to-severe AR (Fisher's exact test, P = 0.007) compared to children with the CC/CT genotypes. There was no significant association between the -509C/T polymorphism and serum TGF-β1 levels (F = 0.389, P = 0.679); however, serum total IgE (F = 4.210, P = 0.018) and ECP (H = 6.297, P = 0.043) levels were found to be significantly associated with the polymorphism.</p><p><b>CONCLUSION</b>The results suggest that the TGFB1 gene polymorphism -509C/T may play a potential role in the severity of persistent AR in childhood.</p>

<p><b>BACKGROUND</b>A satisfactory animal model of breast cancer metastasizing to bone is unavailable. In this study, we used human breast cancer stem-like cells and human bone to build a novel "human-source" model of human breast cancer skeletal metastasis.</p><p><b>METHODS</b>Human breast cancer stem-like cells, the CD44+/CD24-/lower subpopulation, was separated and cultured. Before injection with the stem-like cells, mice were implanted with human bone in the right or left dorsal flanks. Animals in Groups A, B, and C were injected with 1 x 10(5), 1 x 10(6) human breast cancer stem-like cells, and 1 x 10(6) parental MDA-MB-231 cells, respectively. A positive control group (D) without implantation of human bone was also injected with 1 x 10(6) MDA-MB-231 cells. Immunohistochemistry was performed for determination of CD34, CD105, smooth muscle antibody, CD44, CD24, cytokine, CXC chemokine receptor-4 (CXCR4), and osteopontin (OPN). mRNA levels of CD44, CD24, CXCR4, and OPN in bone metastasis tissues were analyzed by real-time quantitative polymerase chain reaction (PCR).</p><p><b>RESULTS</b>Our results demonstrated that cells in implanted human bones of group B, which received 1 x 10(6) cancer stem-like cells, stained strongly positive for CD44, CXCR4, and OPN, whereas those of other groups showed no or minimum staining. Moreover, group B had the highest incidence of human bone metastasis (77.8%, P = 0.0230) and no accompaniment of other tissue metastasis. The real-time PCR showed an increase of CD44, CXCR4, and OPN mRNA in metastatic bone tissues in group B compared with those of groups C and D, however the expression of CD24 mRNA in group B were the lowest.</p><p><b>CONCLUSIONS</b>In the novel "human source" model of breast cancer, breast cancer stem-like cells demonstrated a higher human bone-seeking ability. Its mechanism might be related to the higher expressions of CD44, CXCR4, and OPN, and the lower expression of CD24 in breast cancer stem-like cells.</p>

<p><b>OBJECTIVE</b>This study investigated the effect of hyperbaric oxygenation (HBO) on neural stem cells (NSCs) and myelin in neonatal rats following hypoxic-ischemic brain damage (HIBD) and aimed to explore the possible mechanism of the protective effect of HBO on HIBD.</p><p><b>METHODS</b>Seven-day-old Sprague-Dawley rat pups were randomly assigned into 4 groups: Normal control, HIBD, hyperbaric air (HBA), and HBO groups (n=30 each). The HIBD model was produced by permanent occlusion of the left common carotid artery and 2 hrs hypoxemia exposure (8% O2 at 37 degrees C). HBA and HBO treatment was administered (2 ATA, once daily for 7 days) in the HBA and HBO groups respectively 1 hr after HIBD. BrdU immunohistochemistry was used to detect the NSCs in the sub-ventricle zone (SVZ) of the lateral ventricle and the dentate gyrus (DG) of the hippocampus. The myelin damage was assessed by myelin basic protein (MBP) immunostaining.</p><p><b>RESULTS</b>The BrdU-positive cells in the SVZ and the DG of the ischemic hemisphere in the HIBD group were dramatically decreased compared with those of the Normal control group at 3 weeks post-HIBD (P < 0.01). The HBO treatment resulted in an increase of BrdU-positive cells in the DG from 153.7 +/- 37.0 to 193.7 +/- 38.8 (P < 0.05). The nestin expression in the HIBD and HBA groups was reduced compared with that in the Normal control group. There was no difference in the nestin expression between the HBO and the Normal control groups. Hypoxia-ischemia (HI) led to marked myelin damage at 1 week post-HIBD. HBO or HBA treatment alleviated the damage.</p><p><b>CONCLUSIONS</b>The HBO treatment can result in the proliferation of BrdU-positive cells and alleviate the myelin damage following HIBD in neonatal rats, thereby offering neuroprotectivity against HI insults.</p>

<p><b>OBJECTIVE</b>The overexpression of N-methylpurine DNA glycosylase (MPG) may imbalance the DNA base excision repair (BER) to sensitize tumor cells to current DNA damage chemotherapy. In an effort to improve the efficacy of cancer chemotherapy, we have constructed adenoviral vector of MPG, to study its ability to sensitize human osteosarcoma cell HOS to DNA damage agents.</p><p><b>METHODS</b>The adenoviral infection and MPG expression, as well as enzyme activity were determined by flow cytometry, Western blot, and HEX labeled oligonucleotide-based assay respectively. The cell survival/proliferation was measured using MTS, SRB, and [(3)H] thymidine incorporation assay. Apoptosis cell death was assayed by flow cytometry after treatment using phycoerythin (PE)-conjugated Annexin V and 7-amino-actinomycin (7-AAD).</p><p><b>RESULTS</b>A 10 MOI of recombinant nonreplicating adenovirus was found to infect more than 90% of HOS cells within 24 hours by EGFP fluorescence, in which the MPG overexpression and MPG enzyme activity were also detected. The MPG overexpression HOS cells were significantly more sensitive to the DNA damage agents, including MMS, MNNG, and TMZ, with changes in the IC50 of 6.0, 4.5, and 2.5 fold respectively.</p><p><b>CONCLUSIONS</b>These data establish transient MPG overexpression as a potential therapeutic approach for increasing HOS cellular sensitivity to DNA damage agent chemotherapy.</p>

<p><b>OBJECTIVE</b>To investigate the distribution patterns and proliferative activity of lymphatic vessels in colorectal carcinomas (CRC) and their relationship with tumor metastasis and disease prognosis.</p><p><b>METHODS</b>The microlymphatic density (MLD) and microvascular density in tumoral and non-tumoral areas of 96 cases of CRC were evaluated by immunohistochemistry, using monoclonal antibodies for podoplanin and CD34 respectively. The Ki-67 expression of the lymphatic and blood vessels was detected by double-labeling immunohistochemistry. The relationship between MLD and clinicopathologic features and prognosis was analyzed.</p><p><b>RESULTS</b>The lymph vessels at central and superficia1 portions of CRC often had a reticular architecture with numerous tiny and ill-defined lumina, while those at the tumor borders had large and open lumina. The MLD at tumor borders (51.2 +/- 25.5) was significantly higher than that in normal colorectal mucosa (29.4 +/- 9.0) and other portions of CRC (P < 0.01). The Ki-67 labeling index of the lymphatic lining cells at tumor borders (0.23 +/- 0.17) was significantly higher than that in other portions of CRC (P < 0.05). The MLD significantly correlated with lymphatic involvement by tumor cells, regional lymph node metastasis and distant metastasis (P < 0.01). The 5-year survival rate was also significantly lower in patients with high MLD (P < 0.05).</p><p><b>CONCLUSIONS</b>Neolymphatic vessels are commonly seen in CRC, especially at tumor borders. High MLD at tumor borders is associated with metastasis. The detection of MLD at tumor borders may thus be useful in predicting lymph node metastasis and prognosis in patients with CPC.</p>
